Influence of Stress on Cell Growth and Product Formation Excerpt :

Stresses which arise in bioreactors can influence process performance considerably. Recent molecular biological investigations indicate that stress caused by fluid dynamical effects and extreme values of process variables and toxic substances cause similar responses in the cells. These molecular fundamentals, as well as quantitative evaluation of fluid dynamical stresses and, their effects on microorganisms, animal and plant cells and proteins are treated in this volume.

Plant Tissue Culture Engineering Excerpt :

It is my privilege to contribute the foreword for this unique volume entitled: “Plant Tissue Culture Engineering,” edited by S. Dutta Gupta and Y. Ibaraki. While there have been a number of volumes published regarding the basic methods and applications of plant tissue and cell culture technologies, and even considerable attention provided to bioreactor design, relatively little attention has been afforded to the engineering principles that have emerged as critical contributions to the commercial applications of plant biotechnologies. This volume, “Plant Tissue Culture Engineering,” signals a turning point: the recognition that this specialized field of plant science must be integrated with engineering principles in order to develop efficient, cost effective, and large scale applications of these technologies. I am most impressed with the organization of this volume, and the extensive list of chapters contributed by expert authors from around the world who are leading the emergence of this interdisciplinary enterprise. The editors are to be commended for their skilful crafting of this important volume. The first two parts provide the basic information that is relevant to the field as a whole, the following two parts elaborate on these principles, and the last part elaborates on specific technologies or applications.

Micro and Macroparticle enhanced cultivation of filamentous Lentzea aerocolonigenes for increased rebeccamycin production Excerpt :

The filamentous actinomycete Lentzea aerocolonigenes produces the antitumor antibiotic rebeccamycin. However, the complex morphology of actinomycetes leads to challenges during the cultivation often accompanied by low product titers. In the recent past, the to date low rebeccamycin titers were increased by particle addition to cultivations of L. aerocolonigenes. In this thesis the addition of micro-, macro- and adsorbent particles to cultivations of L. aerocolonigenes were investigated in more detail. Furthermore, the scale-up to a bubble-free bioreactor was conducted. The addition of glass microparticles (x50 = 7.9 μm, 10 g L-1) to shake flask cultivations increased the rebeccamycin titer up to 3.6-fold compared to an unsupplemented approach. Pellet slices showed the incorporation of microparticles. With different surface modifications of the microparticles, specific incorporation patterns of the microparticles appeared. The incorporation of microparticles causes looser and smaller pellets allowing an increased nutrient and oxygen supply in the pellet core. With addition of larger (glass) macroparticles (ø = 0.2 – 2.1 mm, 100 g L-1) mechanical stress was induced on the biopellets. The additional supplementation of 5 g L-1 soy lecithin and glass beads (ø = 969 μm, 100 g L-1) resulted in a rebeccamycin titer of 388 mg L-1, one of the highest rebeccamycin titers ever achieved. For the scale-up of L. aerocolonigenes cultivations a bubble-free membrane aeration system was developed. The tubular membrane aeration system can additionally be pressurized to increase the oxygen transfer. First cultivations of L. aerocolonigenes successfully provided 18 mg L-1 rebeccamycin, a concentration similar to that of unsupplemented shake flask cultivations. XAD adsorbent particles were added to cultivations to facilitate rebeccamycin recovery. However, the XAD particles additionally increased the rebeccamycin titer which was likely to be caused by the adsorption of the rebeccamy

Plant Biotechnology and Molecular Markers Excerpt :

The genesis of the volume, Plant Biotechnology and Molecular Markers, has been the occasion of the retirement of Professor Sant Saran Bhojwani from the Department of Botany, University of Delhi. For Professor Bhojwani, retirement only means relinquishing the chair as being a researcher and a teacher which has always been a way of life to him. Professor Bhojwani has been an ardent practitioner of modern plant biology and areas like Plant Biotechnology and Molecular Breeding have been close to his heart. The book contains original as well as review articles contributed by his admirers and associates who are experts in their area of research. While planning this contributory book our endeavour has been to incorporate articles that cover the entire gamut of Plant Biotechnology, and also applications of Molecular Markers. Besides articles on in vitro fertilization and micropropagation, there are articles on forest tree improvement through genetic engineering. Considering the importance of conservation of our precious natural wealth, one article deals with cryopreservation of plant material. Chapter on molecular marker considers DNA indexing as markers of clonal fidelity of in vitro regenerated plants and prevention against bio-piracy. A couple of write-ups also cover stage-specific gene markers, DNA polymorphism and genetic engineering, including raising of stress tolerant plants to sustain productivity and help in reclamation of degraded land.

Cell and Tissue Reaction Engineering Excerpt :

The completion of the Human Genome Project and the rapid progress in cell bi- ogy and biochemical engineering, are major forces driving the steady increase of approved biotech products, especially biopharmaceuticals, in the market. Today mammalian cell products (“products from cells”), primarily monoclonals, cytokines, recombinant glycoproteins, and, increasingly, vaccines, dominate the biopharmaceutical industry. Moreover, a small number of products consisting of in vitro cultivated cells (“cells as product”) for regenerative medicine have also been introduced in the market. Their efficient production requires comprehensive knowledge of biological as well as biochemical mammalian cell culture fundamentals (e.g., cell characteristics and metabolism, cell line establishment, culture medium optimization) and related engineering principles (e.g., bioreactor design, process scale-up and optimization). In addition, new developments focusing on cell line development, animal-free c- ture media, disposables and the implications of changing processes (multi-purpo- facilities) have to be taken into account. While a number of excellent books treating the basic methods and applications of mammalian cell culture technology have been published, only little attention has been afforded to their engineering aspects. The aim of this book is to make a contribution to closing this gap; it particularly focuses on the interactions between biological and biochemical and engineering principles in processes derived from cell cultures. It is not intended to give a c- prehensive overview of the literature. This has been done extensively elsewhere.

Plant Biotechnology Excerpt :

Biotechnology, is the manipulation of biological organisms to make products that benefit human beings. Biotechnology contributes to such diverse areas as food production, waste disposal, mining and medicine. Plant biotechnology may be defined as the art, science and application of knowledge obtained from the study of life sciences to create technological improvements and change the genetics of plants in order to produce desired characteristics in plant species. This can be accomplished through many different techniques ranging from simply selecting plants with desirable characteristics for propagation, to more complex molecular techniques. Genetic engineering deals with synthesis of artificial gene, repair of gene, combining of DNA from two organism and manipulating the artificial gene together with the recombinant DNA for the improvement of microbes in plants as well as other living being. Genetic engineering opens a totally new dimension for bioprospecting. The search for new genes and their application is the primary objective of the biotech industry. Gene technology now enable humans to integrate revolutionary new properties in to cultivated plants through inter-specific or inter-generic gene transfer which was not possible through classical approach of crop improvement. This book covers all important aspects of practical utility in field of genetic manipulation by different areas of Plant Biotechnology Techniques.
