Genome editing has become the most used tool to produce genetically engineered animals. Specifically in livestock species, the CRISPR/Cas9 system has allowed hundreds of animal models in the last three years to be generated. Despite the high efficiency of this genetic tool, there are still deficiencies of the system that compromise the infinite number of possibilities to induce complex genetic modifications. In vitro studies, as well as intense mouse embryo work paved the way for all the experiments that are currently performed in livestock. However, specific studies in livestock species are required to optimize and explore different possibilities of the system. The sheep is an accepted biomedical model for the scientific community, it also has a comparable size to humans and a well understood physiology that is reflected in the efficient platform for assisted reproductive technologies that are established for this species. The possibility of using sheep as hosts for human organ generation is the main drive of this dissertation. In order to explore this idea, a series of experiments were performed approaching the two critical steps that would be required: generating the “niche” in the host animals and efficiently producing chimeras. We first sought to confirm the possibility of creating a developmental niche by inducing a PDX1-KO using the CRISPR/Cas9 system for generating an apancreatic sheep (Chapter 2). Next, we did a series of experiments to refine the system intending to improve the production of KO sheep using PDX1 as the target gene (Chapter 3). To test the possibility of creating human-sheep chimeras we tried different human iPSCs conditions in sheep embryos. From these studies we found that inhibition of apoptosis increases human chimera contribution in sheep embryos (Chapter 4).
