The Brain and Host Defense Excerpt :

It is now well recognized that the brain, and especially the hypothalamus, plays an important role in the regulation of immune reactions and inflammation. This book aims to review our current state of knowledge of this important field. Key historical findings are presented, and the reciprocal interactions between the brain and the immune system are examined. Particular emphasis is placed on inflammation, a critical host defense reaction that serves as an effector response for both the adaptive and innate immune systems. Mechanisms implicated in brain defense, as well as in more general host defense, are discussed. The regulatory influences of the brain on inflammatory responses are included with particular reference to the role of the hypothalamus, which is also the main director the hormonal regulation of immune/inflammatory. Gender-related differences in immune responsiveness, circadian modulator of immune responses, and evidence that behavioral conditioning (e.g. reward) of immune responses is possible are used as examples to reinforce the notion that the neuroendocrine system exerts a fundamental and complex regulatory influence on the immune system. * Presents timely issues such as immunological aspects of the blood-brain-barrier and the role of inflammatory mediators in the evolution of strokes and degenerative diseases * Includes analysis of the role of the brain in the adaptive responses to disease * Evaluates the argument that further knowledge of the influence of the brain on the immune system will provide new insights to the pathophysiology infectious and autoimmune diseases

Biology of Brain Disorders Excerpt :

Brain disorders, including neurological and neuropsychiatric conditions, represent a challenge for public health systems and society at large. The limited knowledge of their biology hampers the development of diagnostic tools and effective therapeutics. A clear understanding of the mechanisms that underlie the onset and progression of brain disorders is required in order to identify new avenues for therapeutic intervention. Overlapping genetic risk factors across different brain disorders suggest common linkages and pathophysiological mechanisms that underlie brain disorders. Methodological and technological advances are leading to new insights that go beyond traditional hypotheses. Taking account of underlying molecular, cellular and systems biology underlying brain function will play an important role in the classification of brain disorders in future. In this Research Topic, the latest advances in our understanding of biological mechanisms across different brain disorders are presented. The areas covered include developments in neurogenetics, epigenetics, plasticity, glial cell biology, neuroimmune interactions and new technologies associated with the study of brain function. Examples of how understanding of biological mechanisms are translating into research strategies that aim to advance diagnoses and treatment of brain disorders are discussed.

New Insights into Inflammatory Bowel Disease Excerpt :

The incidence of chronic inflammatory and autoimmune diseases is steadily increasing. Inflammatory bowel disease (IBD) is one example of a chronic inflammatory disease, which primarily affects the intestine but may also affect extraintestinal organs. The exact pathogenesis of IBD is currently unknown. However, it is clear that the pathogenesis is complex, involving barrier defects, changes in the intestinal microbiome, and chronic immune activation. This book aims to summarize basic aspects of these complex interactions between barrier function, microbiome, and the immune system. Of note, there is currently no cure for IBD. However, several therapies have evolved in the last years, which are overall able to-at least temporarily-suppress IBD. These therapies and the underlying mechanism are discussed.

Neuro Immune Interactions in Inflammation and Autoimmunity Excerpt :

The nervous system plays an important role in the regulation of immunity and inflammation. On the other hand unbalanced immune responses in inflammatory and autoimmune conditions may have a deleterious impact on neuronal integrity and brain function. Recent studies have characterized neural pathways communicating peripheral inflammatory signals to the CNS, and brain- and spinal cord-derived circuitries controlling various innate and adaptive immune responses and inflammation. A prototypical neural reflex circuit that regulates immunity and inflammation is the vagus nerve-based “inflammatory reflex”. Ongoing research has revealed cellular and molecular mechanisms underlying these neural circuits and indicated new therapeutic approaches in inflammatory and autoimmune disorders. Pharmacological and bioelectronic modulation of neural circuitry has been successfully explored in preclinical settings of sepsis, arthritis, inflammatory bowel disease, obesity-driven disorders, diabetes and other diseases. These studies paved the way to successful clinical trials with bioelectronic neuronal modulation in rheumatoid arthritis and inflammatory bowel disease. Dysregulated release of cytokines and other inflammatory molecules may have a severe impact on brain function. Brain inflammation (neuroinflammation), imbalances in brain neuronal integrity and neurotransmitter systems, and cognitive impairment are characteristic features of post-operative conditions, sepsis, liver diseases, diabetes and other disorders characterized by immune and metabolic dysregulation. Derangements in cytokine release also play a pivotal role in depression. Characteristic brain reactive antibodies in autoimmune conditions, including systemic lupus erythematosus and neuromyelitis optica, significantly contribute to brain pathology and cognitive impairment. These studies, and the simultaneous characterization of neuro-protective cytokines, identified new therapeutic approaches for treating neurological

Understanding Crohn s Disease Immunity Genes and Microbes Excerpt :

Crohn's disease (CD) is a chronic, relapsing, inflammatory bowel disease resulting in considerable morbidity and reduced quality of life. Although still under intense debate, CD seems to result from an enhanced and uncontrolled immune response to the gut microbiota. CD is thought to be multifactorial depending on genetic and environmental determinants. In recent years, nearly 100 single nucleotide polymorphisms (SNPs) were associated with increased risk of developing CD (some of the SNPs also associated with susceptibility to ulcerative colitis, another type of IBD). These SNPs are mostly located in genes involved in innate and adaptive immunity mechanisms, such as autophagy, expression of pattern-recognition receptors and citokine signaling. Epigenetics is also probably playing a role in CD susceptibility, as it is sensitive to environmental conditions and may mediate gene-environment interactions. Environmental factors possibly involved in CD development include diet, gut microbiota composition and infection with specific pathogens, of which the most consistently associated to CD are Mycobacterium avium subsp. paratuberculosis and adherent-invasive Escherichia coli. This Topic aimed at bringing together contributions covering different genetic, epigenetic, immunological and microbial processes involved in the development of CD, helping to drive forward the understanding of CD immunopahtology.
