The Chemistry and Biology of Nitroxyl HNO Excerpt :

The Chemistry and Biology of Nitroxyl (HNO) provides first-of-its-kind coverage of the intriguing biologically active molecule called nitroxyl, or azanone per IUPAC nomenclature, which has been traditionally elusive due to its intrinsically high reactivity. This useful resource provides the scientific basis to understand the chemistry, biology, and technical aspects needed to deal with HNO. Building on two decades of nitric oxide and nitroxyl research, the editors and authors have created an indispensable guide for investigators across a wide variety of areas of chemistry (inorganic, organic, organometallic, biochemistry, physical, and analytical); biology (molecular, cellular, physiological, and enzymology); pharmacy; and medicine. This book begins by exploring the unique molecule’s structure and reactivity, including important reactions with small molecules, thiols, porphyrins, and key proteins, before discussing chemical and biological sources of nitroxyl. Advanced chapters discuss methods for both trapping and detecting nitroxyl by spectroscopy, electrochemistry, and fluorescent inorganic cellular probing. Expanding on the compound’s foundational chemistry, this book then explores its molecular physiology to offer insight into its biological implications, pharmacological effects, and practical issues. Presents the first book on HNO (nitroxyl or azanone), an increasingly important molecule in biochemistry and pharmaceutical research Provides a valuable coverage of HNO’s chemical structure and significant reactions, including practical guidance on working with this highly reactive molecule Contains high quality content from recognized experts in both industry and academia

Free Nitroxyl Radicals Excerpt :

In the short time since the first nitroxyl radical was obtained in 1959, a new branch of chemical science has arisen and taken shape-the chemistry of stable nitroxyl radicals. The present book was written by E. G. Rozantsev, one of the pioneers in this field and a prominent specialist on stable radicals at the Division of Chemical and Biological Processes of the Institute of Chemical Physics, Academy of Sciences of the USSR. His numerous papers have contributed greatly to the present situation, wherein nitroxyl radicals have acquired unusually wide popularity, including the fields of biology, medicine, chemis try, physics, biophysics, and instrument construction. A clear illustration of the astonishingly vigorous development of this new field of chemistry can be given by the enormous flow of information on the synthesis and use of nitroxyl radicals. There is no doubt that this monograph, which in part generalizes the results of many workers studying these radicals, will be received with interest by specialists working in this field. The author has not attempted to give an exhaustive account of the material. His aim is to introduce the reader to this new field and to show the wide possibilities for using radicals in scientific experiment. The voluminous bibliography, including many papers by the author himself which may not be well enough known to the American reader, will undoubtedly contribute to the usefulness of the monograph.

Synthetic Explorations in the Pursuit of a Rapid Photoactivatable Nitroxyl Donor Excerpt :

Nitroxyl (HNO) is a biologically relevant small molecule with considerable clinical promise for the treatment of heart failure. However, studies of the chemistry and biology of nitroxyl are hampered by its instability in aqueous solution. To aid in biological and chemical studies, nitroxyl donors (molecules that degrade to release HNO) have been synthesized which utilize a 3-hydroxy-2-naphthyl methyl (HNM) OH protecting group. Our group0́9s focus is on the synthesis of nitroxyl donors that rapidly release HNO under physiological conditions through photoactivation. First generation HNO donors released HNO upon photoactivation, but competition was also observed from a redox side reaction. In this study, we are probing the impact of a methyl substituent at the benzylic position on these competing processes. The synthesis and photochemical anaylsis of such a donor are herein described.

Oxidation and Antioxidants in Organic Chemistry and Biology Excerpt :

Providing a comprehensive review of reactions of oxidation for different classes of organic compounds and polymers, and biological processes mediated by free radicals, Oxidation and Antioxidants in Organic Chemistry and Biology puts the data and bibliographical information you need into one easy-to-use resource. You will find up-to-date information about mechanisms of action of antioxidants, their reactivity, reactions of intermediates, synergism, and antioxidants with cyclic mechanism action. Supplying useful, quantitative data in tables that make the information easy to find, the authors highlight the peculiarities of mechanisms involved in the oxidation of hydrocarbons, polymers, and different organic compounds. The book provides tabulated values of strengths of C-H bonds of oxygen-containing compounds; of O-H bonds of hydroperoxides, alcohols, and acids; and of attacked antioxidant bonds. The authors collect and discuss over 3000 rate constants of different reactions of peroxyl radicals in oxidation and co-oxidation. They describe a new semiempiric theory of reactivity of reactants in elementary oxidative steps and the algorithm of calculation of activation energies, rate constants, and geometrical parameters of the transition states of free radical reactions. After elucidating the chemistry and kinetics of antioxidant action, the book covers oxidative processes that occur in biological systems.

This book covers the basic theory and techniques, as well as various applications of pulsed electron–electron double resonance (PELDOR or DEER). This electron paramagnetic resonance technique is able to measure the distances and the distribution of distances between electron spins in the 1.5–15 nanometer scale; to determine the geometry of spin-labeled molecules; to estimate the number of interacting spins in spin clusters; and to characterize the spatial distribution of paramagnetic centers. As a result, PELDOR is now a popular method in EPR spectroscopy, particularly in the context of biologically important systems and soft matter and is also applied to problems in physical chemistry, biochemistry, polymers, soft matter and materials. Enabling readers to gain an understanding of the fundamentals of the PELDOR methods and an appreciation of the opportunities PELDOR provides, the book helps readers solve their own physical and biochemical problems.

Nitrosyl Complexes in Inorganic Chemistry Biochemistry and Medicine II Excerpt :

The series Structure and Bonding publishes critical reviews on topics of research concerned with chemical structure and bonding. The scope of the series spans the entire Periodic Table and addresses structure and bonding issues associated with all of the elements. It also focuses attention on new and developing areas of modern structural and theoretical chemistry such as nanostructures, molecular electronics, designed molecular solids, surfaces, metal clusters and supramolecular structures. Physical and spectroscopic techniques used to determine, examine and model structures fall within the purview of Structure and Bonding to the extent that the focus is on the scientific results obtained and not on specialist information concerning the techniques themselves. Issues associated with the development of bonding models and generalizations that illuminate the reactivity pathways and rates of chemical processes are also relevant. The individual volumes in the series are thematic. The goal of each volume is to give the reader, whether at a university or in industry, a comprehensive overview of an area where new insights are emerging that are of interest to a larger scientific audience. Thus each review within the volume critically surveys one aspect of that topic and places it within the context of the volume as a whole. The most significant developments of the last 5 to 10 years should be presented using selected examples to illustrate the principles discussed. A description of the physical basis of the experimental techniques that have been used to provide the primary data may also be appropriate, if it has not been covered in detail elsewhere. The coverage need not be exhaustive in data, but should rather be conceptual, concentrating on the new principles being developed that will allow the reader, who is not a specialist in the area covered, to understand the data presented. Discussion of possible future research directions in the area is welcomed. Review articles for
