"AbstractThe Hippo signaling pathway plays an important role during development, tissue regeneration and tumorigenesis. The Hippo-regulated transcriptome in the gut epithelium includes a broad range of secreted factors including the pro-survival factor clusterin (Clu). We hypothesized that Clu-expressing cells acquire Yap-dependent intestinal stem cell (ISC) activity in response to injury. Using single molecule fluorescent in situ hybridization (sm-FISH), we showed that Clu expressing cells are distinct from Lgr5+ intestinal stem cells. Under normal circumstances, Clu was expressed at a very low levels by epithelial cells located at the bottom of the crypts. However, in contrast to Lgr5, whole body gamma-irradiation triggered a strong upregulation of Clu in the crypt epithelium. Furthermore, during the regenerative phase following irradiation, Clu and Lgr5-expressing cells were found to be mutually exclusive from one another. The potential function of Clu during crypt regeneration was examined by isolating and culturing the crypt epithelium from CluWT and CluKO mice as 3D organoids in Matrigel. To identify differentially expressed genes in control and Clu-deficient regenerating crypts, we isolated RNA from organoid cultures and performed qPCR experiments at multiple time points for our selected genes. To investigate the function of Hippo signaling pathway in an infectious model we examined the role of Yap following intestinal infection by Heligmosomoides polygyrus bakeri (Hpb). Recent studies showed that helminth-associated crypts acquire an interferon-gamma (Ifng)-mediated fetal reprograming during Hpb infection. To determine whether Yap-induced Clu+ stem cells were activated during Hpb infection, we performed lineage tracing experiments with CluCreERT; LSL-tdTomato mice. Several chapters focus on the importance of these genes in leukemia research, where RUNX1 and CBFB are frequently affected by chromosomal translocations that generate fusion oncoproteins, while recent studies suggest wider roles for RUNX modulation in solid cancers.
